Vital imaging of glomeruli in the mouse olfactory bulb.

چکیده
We have monitored the pattern of identified glomeruli in the olfactory bulbs of newborn, juvenile, and adult mice over intervals of several hours to several weeks. Our purpose was to assess the development and stability of these complex units in the mammalian brain. Glomeruli can be observed by vital fluorescent staining and laser-scanning confocal microscopy without causing acute or long-term damage to brain tissue. Repeated observation of bulbs in the same animals between birth and 3 weeks of age showed that this region of the brain develops by progressive addition of these units to the original population. This increment occurs by the genesis of smaller new glomeruli between larger existing ones; no elimination of glomeruli was observed during this process. Finally, no addition (or loss) of glomeruli occurred in adult animals over a 2 week interval; once established, the number, size, and pattern of glomeruli are evidently stable.

Selective imaging of presynaptic activity in the mouse olfactory bulb shows concentration and structure dependence of odor responses in identified glomeruli.
More chemicals can be smelled than there are olfactory receptors for them, necessitating a combinatorial representation by somewhat broadly tuned receptors. To understand the perception of odor quality and concentration, it is essential to establish the nature of the receptor repertoires that are activated by particular odorants at particular concentrations. We have taken advantage of the one-t...
متن کاملSymmetry, stereotypy, and topography of odorant representations in mouse olfactory bulbs.
The molecular basis of vertebrate odorant representations has been derived extensively from mice. The functional correlates of these molecular features were visualized using optical imaging of intrinsic signals in mouse olfactory bulbs. Single odorants activated clusters of glomeruli in consistent, restricted portions of the bulb. Patterns of activated glomeruli were clearly bilaterally symmetr...
متن کاملIn Vivo Imaging of Neuronal Activity by Targeted Expression of a Genetically Encoded Probe in the Mouse
Genetically encoded probes show great promise in permitting functional imaging of specified neuronal populations in the intact nervous system, yet their in vivo application has been limited. Here, we have targeted expression of synapto-pHluorin, a pH-sensitive protein that reports synaptic vesicle fusion, to olfactory sensory neurons in mouse. Synapto-pHluorin selectively labeled presynaptic te...
متن کاملWavelet-based multi-resolution statistics for optical imaging signals: Application to automated detection of odour activated glomeruli in the mouse olfactory bulb.
Optical imaging techniques offer powerful solutions to capture brain networks processing in animals, especially when activity is distributed in functionally distinct spatial domains. Despite the progress in imaging techniques, the standard analysis procedures and statistical assessments for this type of data are still limited. In this paper, we perform two in vivo non-invasive optical recording...
متن کاملPostnatal refinement of functional odor maps on the surface of the mouse olfactory bulb
Newborn mice rely on olfactory information for survival, but the nature of odor representation in the neonatal olfactory bulb is not well understood. I compared the spatial pattern of odor-evoked responses in 1 to 11 week old mice by imaging the dorsal surface of olfactory bulbs. By using a large panel (~100) of chemically diverse odorants I was able to assign functional tags to individual glom...
